After talking to Ada and Shez about it, I've decided to run one. Knowledge of the game world or the mechanics aren't necessary. But a few basic setting notes might be useful. L5R is set in Rokugan, an ancient and tradition-bound empire that's a lot like Japan or China. The land is divided between eight Great Clans, each of which takes their name from an animal that represents their philosophies and attitudes on life. The Emperor rules by divine mandate, and by balancing the power and ambitions of the clans against each other. The noble class are the Samurai - they are trained in ancestral techniques to serve as bushi (warriors), shugenja (magic-using priests) or courtiers. To the south is the Shadowlands, which is a direct link to Jigoku (hell). From the Shadowlands, supernatural evil comes forward constantly to attack the Empire.

I also need to know which game system you all prefer to use. D10 is the official l5r system, designed specifically for the world, and obviously it's the best fit. d20 had a conversion, and most people are familiar with d20, but it is an inferior rules-set for l5r. Or there's the SPAM system, which is my homebrew system. Incredibly rules-light, focused on story over rules, but is still being polished.
